Nació el 26 de mayo de 1907 en Winterset, Iowa (Estados Unidos).
Hijo de Clyde Morrison y de Mary Alberta Brown. Siendo un niño su familia se trasladó aLancaster, en el desierto del Mojave debido a una afección pulmonar del padre buscaban un clima más cálido. Se hicieron cargo de un pequeño rancho, donde el pequeño Marion aprende a montar a caballo con soltura. Posteriormente se trasladaron a Glendole, en California, una localidad más cercana a Los Angeles. El padre se emplea en una farmacia y él, con once años -todavía en la escuela- empieza a trabajar repartiendo periódicos y las recetas de su padre aficionándose al cine donde trabaja los sábados en una sala del pueblo.
Sobresalió como deportista en la universidad de Southern California, aunque dejó el deporte para dedicarse al cine, donde se inició como atrezzista en la Fox.
LA GRAN JORNADA
Aparece en pequeños papeles y aparece por primera vez en los créditos de una película con el nombre de "Duke" Morrison (Words and Music, 1929). Pero Walsh es quien, en La gran jornada, un año después lo lanzará (y con el nombre que va a conservar) encarnando a un seductor peinado con brillantina, recién salido de las manos del sastre. Siguieron otros filmes de poca importancia, protagonizó películas de serie B para la Monogram o la Republic y algunos seriales, como The three musketeers, o The Hurricane Express, doce episodios de acción, hasta que en 1939, con La diligencia, de John Ford, se inició una de las más brillantes carreras de la historia del cine.
LA DILIGENCIA
FORT APACHE
Fue una gran estrella durante 40 años, sobresaliendo en películas como Fort Apache (1948), El hombre tranquilo (1952) o Centauros del desierto (1953), dirigidas también por Ford, El Álamo(1960) o Boinas verdes (1968), producidas y dirigidas por él mismo o Valor de ley (1969), de Henry Hathaway. Wayne brilló también en Amor a reacción, Escrito bajo el sol, Hatari, o en El fabuloso mundo del circo. Ganó un Óscar al mejor actor por su papel en la película Valor de ley. Apareció por última vez en la gran pantalla en El último pistolero (1976), de Don Siegel.
Intervino en 157 películas, bajo la dirección de los realizadores más prestigiosos de Hollywood, aunque su carrera estuvo marcada por los 20 largometrajes que rodó con John Ford.
Contrajo matrimonio con Josephine Alicia Sánchez de la que se divorció en 1945, después con Esperanza Baur, de quien se divorció en 1954 y finalmente con Pilar Palette. Con Josephine tuvo cuatro hijos, y tres con Pilar.
En 1963, se le detectó un cáncer de pulmón, y en 1978 sufre una operación a corazón abierto. Después es operado de cáncer en los intestinos y en el estómago.
John Wayne murió rodeado de sus siete hijos el 11 de junio de 1979 en el Centro Médico Universitario de Los Angeles a los 72 años de edad, víctima de un cáncer generalizado. Fue enterrado en el Pacific View Memorial Park de Newport, con el epitafio en castellano: " Feo, fuerte y formal ".
